The changes in DA concentration of the dorsal striatum after the acute administration of saline or β-PEA were measured using a DA ELISA kit (#KA1887, Abnova, Taipei, Taiwan). According to the manual, standards, controls, and samples (protein lysates of the dorsal striatum) were subjected to extraction and acylation assays. The acylated standards, controls, and samples were pipetted into the appropriate wells of the DA microtiter strips and incubated with DA antiserum for 30 min at room temperature on a shaker. The conjugate was added to all wells and incubated for 15 min, and the wells were washed. After 3 washes for 10 min, the stop solution was added into the wells, and then the absorbance was measured at 450 nm in a GloMax microplate multimode reader (Promega, Madison, WI, USA).

Depending upon stereochemistry, norephedrine is the β-OH analog of amphetamine whereas ephedrine is the β-OH analog of methamphetamine; the other two optical isomers are referred to using the prefix “pseudo” (Figure 11). The naturally-occurring (-)ephedrine (53), the most potent of the phenylpropanolamines as a central stimulant, is several times less potent than amphetamine or methamphetamine. Stimulus generalization was demonstrated to be time-dependent, with the phenolic compounds having a longer onset of action time (decreased ability of the agents to penetrate the BBB?). However, there was some concern that 2-DM-DOM and 5-DM-DOM might undergo further O-demethylation in vivo to a hydroquinone. Before leaving the topic of fundus 5-HT receptors, it is worth mentioning that attempts were made to determine whether peripheral rat fundus 5-HT receptors bore any similarity to brain 5-HT1 or 5-HT2 receptors.

Phenethylamines
Monoamine oxidase (MAO) supplements are another popular substance used by depressed patients to try and combat the nasty side effects of phenethylamine. Like most supplements, there is good, reliable information out there concerning the efficacy of these products in helping to treat phenylamine-induced behavior disorders and the like. However, it is also important for anyone considering MAO products to know and understand that even though the substances themselves are relatively safe, any MAO product, even those sold over-the-counter, can produce a dangerous build-up of potentially harmful conditions if used improperly. Although it’s mainly used as a neurotransmitter in the human brain, phenethylamine can also be produced in other organisms. As we look to the future, it’s crucial to encourage further research into PEA and its effects on brain function. There is still much to learn about its long-term effects, optimal dosages, and potential interactions with other substances.
Finally, 100 µM DMBMPP resulted in large aggregate forms of various size and shape, indicating that DMBMPP inhibits proper microtubule polymerization. General Pharmacology Of Phenethylamines

One last condition related to phenylethylamine and MAO is dyme dystrophy, or “white hand”, a rare condition in which the body lacks the proper amount or activity of a specific set of neurotransmitters known as norepinephrine and epinephrine. Because this condition frequently accompanies high levels of anxiety or depression, it can be used to test the effects of low doses of phenethylamine on these neurotransmitters. Unfortunately, much of the evidence surrounding the use of this substance for these purposes comes from the work of autistic individuals, who are most at risk for developing type dystrophy as a result of prolonged phenylamine use. There is, however, sufficient evidence to suggest that there may be some benefit to individuals with other mental disorders, particularly if they exhibit a chronic lack of concentration or poor impulse control. For example, individuals with attention deficit disorder, attention deficit hyperactivity disorder, depression, and other mood disorders have all been shown to be at a higher risk of developing dymestrophy. The degree of effect on dopamine neurotransmission and duration of action appear to help guide understanding of habituation.

It was the latter studies that opened new avenues of pursuit to us (e.g. 5-HT1A, 5-HT6, and other receptors). To maintain exponential growth and considering that the time required to complete the cell cycle is 13–14 h, the cultures were divided every three days in fresh medium, and the cell density did not exceed the critical value of 9 × 105 cells/mL. MDMA did not show a cytotoxic and cytostatic effect at all concentrations tested and did not increase the MNi frequency in any case. It’s particularly helpful if you take Phenylethylamine prior to a stressful situation, workout, or physically demanding job. Do not use Phenylethylamine (PEA) if you are using a prescription MAOI like Marplan, Nardil, Azilect or Parnate, or have used one in the last 14 days. Dosing more than recommended is NOT a good idea because you’ll likely feel jittery, irritable, get a headache, feel nausea, and very possibly force your heart rate to dangerous levels.
Subsequent studies showed that α-ET produced MDMA-like stimulus effects (Table 8). It was looking more and more like α-ET was an indolealkylamine counterpart of MDA. As already alluded to,74 synthetic cathinones don’t represent a pharmacologically homo-geneous group of agents. Depending upon terminal amine and aryl substituents, stereochemistry, and the nature of the α side chain, these agents can act as releasing agents or re-uptake inhibitors at DAT, NET, and/or SERT.
Ortho or meta positions enhanced activity, whereas the para substitution reduced it. One of the best derivatives was 140, which was compared with its tryptamine congener 141, less potent overall in the 5-HT2 assays performed (Figure 17c). Based on these catecholamines, several studies were performed to investigate the effect of chirality, further functionalization, and activity on different derivatives 25,26,27,28,29,30,31. This later triggered the elaboration of many derivatives conserving the 2-phenylethyl moiety, which have been frequently used in the context of medicinal chemistry as tool compounds (Figure 5, Table 1). Pharmaceutical drugs that are substituted phenethylamines include phenelzine, phenformin, and fanetizole, among many others. The tubulin polymerization Assay (BK006P) was purchased from Cytoskeleton Inc. (Denver, CO, USA) and it was used with the purpose to measure the optical density of each sample over time using a spectrophotometer at a temperature of 37 °C at a wavelength of 355 nm on 96 well-plates.
Compound 9, which has a chiral center, can exist as (R)- and (S)-isomers; compound 28, which has two chiral centers was represented as (R, R)-, (R, S)-, (S, R)-, and (S, S)-isomers. The fluorescence intensity of 2′,7′-dichlorofluorescin (DCF) (which forms in cells in the presence of ROS) recorded in treated cultures was normalized on those recorded in the untreated control cultures, accounted equal to 1 and expressed as a ROS fold increase. The MNi frequencies (number of MNi/10,000 nuclei) recorded in treated cultures were normalized on those recorded in the untreated control cultures, accounted equal to 1 and expressed as MNi frequency fold increase. In addition, to make the genotoxicity test reliable, it is necessary to check the cellular proliferation in order to verify that a sufficient number of cells has undergone mitosis and so transmitted the genetic damage suffered to the daughter cells. For this purpose, the OECD recommends the measurement of the Relative Population Doubling (RPD) to estimate the cytostasis and, analogously to the cytotoxicity, establishes a threshold at most equal to 55 ± 5% 45.
Among the characteristics of depression that were summarized in a review article (Voinov et al., 2013), women are 50% more likely to be affected by depression than men and depression can shorten a person’s life by years (Voinov et al., 2013). Training doses were 1.0, 1.0, and 0.5 mg/kg for S(+)AMPH, S(+)METH, and S(-)MCAT, respectively. 5-HT2 receptor agonists 26, 27, 29–31, 40, 41, and the 5-HT2 receptor antagonist M (28). DO NOT make the mistake of buying Phenylalanine instead of Phenylethylamine (PEA). PEA has a half-life of 5 – 10 minutes.xvii But the effects of PEA can be extended by using it with a MAO-B inhibitor. PEA instead only increase the amount of neurotransmitters that get released when a neuron is stimulated by receiving an impulse from a neighboring neuron.
